In Guitar Hero 5 kun je een eigen band vormen, waarbij je zelf bepaalt in welke bezetting dat is. Als je wilt kun je optreden met vier zangeressen, met drie drummers en een gitarist of gewoon twee zangers en gitaristen. Bovendien kun je tijdens het spelen op elk gewenst moment jouw instrument uitzetten en overstappen op een andere. Ook kun je terwijl je speelt een vriend uitnodigen of de moeilijkheidsgraad aanpassen, zonder dat het nummer opnieuw moet worden gestart.

Guitar Hero 5 bevat 85 nummers van bekende artiesten als The Rolling Stones, The White Stripes, Kings of Leon, Tom Petty, Bob Dylan, Johnny Cash, Santana, Vampire Weekend, Beastie Boys, Coldplay, Duran Duran, Gorillaz, Jimmy Eat World, Muse en The Raconteurs.
